Despite a multi length variant approach and an intensive screening campaign, the kinase domain of Polo-like kinase-1 initially resisted all crystallization efforts. Crystals were finally obtained via co-crystallization with a Plk1-specific DARPin developed for this purpose, and, in an alternative approach, via surface entropy reduction mutations. The resulting structures show that both approaches probably succeeded by modifying the same surface patch.
